Rhapontic Rhubarb
Rheum rhaponticum root extract (ERβ-selective)
What it is
Rhapontic rhubarb root contains a family of stilbene glycosides — rhaponticin, desoxyrhapontigenin, and their aglycones — that act as selective estrogen receptor beta (ERβ) agonists. ERβ receptors are distributed throughout the brain, bone, and vascular system. Activating them reduces vasomotor symptoms without the systemic estrogen effects associated with HRT. This is the only non-hormonal botanical mechanism confirmed to target the actual signaling deficit behind hot flashes and night sweats.
Clinical evidence
Twelve randomized, placebo-controlled clinical trials. The landmark Heger study (2006, Menopause journal, 109 women, 12 weeks) documented a 68% reduction in hot flash frequency. A 48-week follow-up confirmed sustained benefit with no detectable uterine stimulation. Safety established up to two years of continuous use.
Why we chose this source
Dosed at 200 mg — the full clinical dose, five times the historical 40 mg used in earlier European studies. ERβ-selective standardization identity-verified by HPLC. Produced under pharmaceutical-grade cGMP conditions. The extract designation ERr 731 refers to the specific standardized fraction used across all twelve published trials.
Berberine HCL
Berberis aristata root extract (97% berberine HCL)
What it is
Berberine is a plant alkaloid extracted from Berberis aristata and related species. It activates AMPK — the enzyme described by researchers as a metabolic master switch — improving insulin sensitivity, glucose uptake, and lipid metabolism. During peri-menopause, the decline of estradiol disrupts these same metabolic pathways, which is why abdominal weight accumulates even in women who have not changed their diet or exercise habits.
Clinical evidence
Harvard Medical School research validated berberine's effect on glucose metabolism and insulin sensitivity. Multiple randomized controlled trials show berberine at 1,000 mg daily performs comparably to metformin on fasting blood glucose and HbA1c. A 2022 meta-analysis in Frontiers in Pharmacology confirmed significant reductions in body weight, BMI, and waist circumference.
Why we chose this source
The HCL salt form, standardized to 97% purity — the most bioavailable form and the one used in virtually all published clinical trials. 1,000 mg is the full pharmaceutical dose. We do not under-dose.
Ashwagandha root extract
Withania somnifera root extract (5% withanolides minimum)
What it is
Ashwagandha is one of the most researched adaptogens in the world, used in Ayurvedic medicine for over 3,000 years. The withanolides — bioactive steroidal lactones concentrated in the root — modulate the HPA axis and reduce cortisol output under chronic stress. During peri-menopause, declining estradiol leaves the stress response more exposed: the same stressors hit harder, sleep becomes fragmented, and anxiety can appear seemingly from nowhere.
Clinical evidence
Published clinical trials on standardized ashwagandha (5% withanolides) document significant reductions in serum cortisol, perceived stress scores, and anxiety, alongside measurable improvements in sleep quality and cognitive performance. A double-blind RCT (Indian Journal of Psychological Medicine) reported cortisol reduction and improved sleep onset versus placebo at comparable doses over 60 days.
Why we chose this source
Root-only extraction standardized to a minimum of 5% withanolides — the active fraction validated in clinical research. Root-only sourcing delivers consistent withanolide concentration without the variable alkaloid profile of whole-plant extracts.
Magnesium Bisglycinate Chelate
Magnesium bis-glycinate chelate (TRAACS grade)
What it is
Magnesium is a cofactor in more than 300 enzymatic reactions — every one involved in sleep onset, muscle relaxation, and nervous system regulation. An estimated 50% of U.S. adults consume less magnesium than the recommended daily intake, and peri-menopausal hormonal shifts accelerate this depletion. Bisglycinate chelate — in which magnesium is bound to two glycine molecules — avoids the GI distress of oxide and citrate forms and crosses the blood-brain barrier for direct central nervous system effects.
Clinical evidence
Meta-analyses consistently show magnesium supplementation improves sleep quality, reduces muscle cramp frequency, and lowers cortisol. A randomized trial in the Journal of Research in Medical Sciences found magnesium supplementation significantly improved insomnia severity scores in adults over 46.
Why we chose this source
TRAACS-grade bisglycinate chelate — the amino acid chelation specification that ensures the mineral is fully bound to its carrier molecule, resisting dissociation in the GI tract. Not the oxide or carbonate forms found in cheap multivitamins, which pass largely unabsorbed.
Black Cohosh root extract
Cimicifuga racemosa root extract (2.5% triterpene glycosides)
What it is
Black cohosh works through non-estrogenic pathways — modulating serotonergic and dopaminergic signaling rather than acting on estrogen receptors — which is why it does not raise the same concerns as phytoestrogens in women with hormone-sensitive histories. It was the subject of the most extensive long-term botanical research program in peri-menopause history, known as the Remifemin protocol, conducted in Germany over several decades.
Clinical evidence
Germany's Commission E approved black cohosh for menopausal symptom management in 1989. Multiple randomized trials confirm meaningful reductions in hot flash frequency and intensity. A systematic review of 16 trials found consistent improvement in vasomotor symptoms with a strong safety profile.
Why we chose this source
Root extract standardized to 2.5% triterpene glycosides — the active markers validated in published trials. Identity-verified by HPLC. Sourced from sustainably managed wild-crafted and cultivated Cimicifuga racemosa.
Saffron extract
Crocus sativus stigma extract (3.5% safranal & crocins minimum)
What it is
Saffron is among the most studied botanicals for emotional wellbeing. Its active compounds — crocins and safranal — modulate serotonin and dopamine reuptake without the side-effect profile of pharmaceutical SSRIs. During peri-menopause, fluctuating estradiol directly disrupts serotonergic signaling, which explains the new-onset anxiety and mood dysregulation that many women experience for the first time in their 40s.
Clinical evidence
Eight published randomized controlled trials on mood and anxiety. A trial specifically in peri-menopausal women showed a 33% reduction in anxiety scores versus 13% in placebo after 12 weeks. Separate trials have demonstrated efficacy comparable to 20 mg fluoxetine for mild-to-moderate depressive symptoms — without the sexual side effects.
Why we chose this source
Stigma-only extraction — the part of the Crocus sativus flower that concentrates the active bioactives. Standardized to a minimum of 3.5% safranal and crocins by HPLC. Saffron is one of the most adulterated ingredients in the supplement industry; identity testing on every batch is non-negotiable.
Vitamin B6 (P5P)
Pyridoxal-5-Phosphate — active coenzyme form
What it is
Vitamin B6 in its P5P form is the active coenzyme the body uses directly — no liver conversion required. It acts as a cofactor in the synthesis of serotonin, dopamine, and GABA simultaneously, placing it upstream of mood, sleep onset, and stress response at once. P5P is also essential for progesterone metabolism, making it directly relevant during peri-menopause when progesterone levels fluctuate most.
Clinical evidence
P5P achieves substantially higher plasma concentrations than pyridoxine HCl, particularly in women over 40 in whom hepatic conversion becomes less reliable. Published European clinical nutrition protocols use P5P specifically for hormonal balance and mood support in peri-menopausal women.
Why we chose this source
Pyridoxal-5-Phosphate — the bioactive coenzyme form, not the inactive pyridoxine HCl found in most supplements. At 20 mg per serving, the dose is clinically meaningful without approaching the sensory neuropathy threshold associated with very high-dose pyridoxine supplementation.
Vitamin K2 MK-7
Menaquinone-7 — natto fermentation (trans-isomer >99%)
What it is
Peri-menopause is the period of most accelerated bone loss in a woman's life. Vitamin K2 MK-7 activates osteocalcin — the protein that binds calcium into bone matrix — and activates matrix GLA protein, which prevents calcium from depositing in arterial walls. It does not increase calcium absorption (that is vitamin D3's role). It governs where absorbed calcium goes. Taking calcium without adequate K2 can be actively counterproductive for cardiovascular health.
Clinical evidence
The Knapen 2013 three-year randomized trial demonstrated MK-7 at 180 mcg significantly improved vertebral bone strength in post-menopausal women. Multiple subsequent studies confirm K2 MK-7's role in bone mineral density and arterial calcification reduction.
Why we chose this source
Produced via natural fermentation of natto (Bacillus subtilis natto) — the traditional Japanese process that yields the biologically active MK-7 form. Trans-isomer purity >99%. This matters: cis-isomers of MK-7 lack biological activity and are a common adulterant in low-quality K2 ingredients.
